Set in 1988, Fear Street: Prom Queen primarily focuses on Shadyside High School. It's Senior Prom time, and things are getting heated up between the candidates. This time, six young women are vouching for the crown, and one of them is Lori Granger (played by India Fowler). A shy girl who doesn't talk to everyone and doesn't want to be in the spotlight. However, her life has been quite tumultuous. Her mother had been accused of killing her father, and that was one of the reasons why her mother could never go to the prom. Although the police couldn't find any evidence against Lori's mother, many believed that it was indeed she who killed her husband. But this time, Lori took a chance and tried to do something bold by nominating herself for the Prom Queen.

Her main competitors are a group of girls called "Wolfpack." The group is led by Tiffany Falconer (played by Fina Strazza, known for her role in Paper Girls), a typical mean girl who wants to be the Prom Queen by any means. Just like any other bully, she makes Lori's life miserable by talking about her mother. Her group of girls includes Melissa (played by Ella Rubin), Debbie (played by Rebecca Ablack), and Linda (played by Ilan O'Driscoll). They are all running for the title of Prom Queen and want Tiffany to win. The entire group keeps picking on Lori, and when it is revealed that she wants to be a writer, Tiffany and Debbie make fun of her by talking about her mother. However, Lori is not alone and has a best friend in the form of Megan (played by Suzanna Son), a stoner girl who doesn't have many friends. She makes sure that Lori is not alone and shocks everyone by chopping off her own hand in the middle of the class. But it turns out to be a prank to scare Tiffany and her group.

There's another girl who is running for the Prom Queen, and her name is Christy (played by Ariana Greenblatt). She acts as a little drug dealer who provides drugs to students. After providing drugs to some students, she was walking back home when she was attacked by a masked man wearing a red dress which looked like a raincoat. He kills her brutally with an axe and eliminates her from the book, where the killer had all the photos of the Prom Queen candidates. What happened next was brutal and filled with anxiety.

Christy gets eliminated from the competition for not being able to show up for the trials. Meanwhile, Tiffany does everything to make Lori realize she doesn't stand a chance against her. As a result, she shouldn't continue and forfeit. However, Lori is not the only one who is giving her competition. One of the girls from the Wolfpack, Linda, is actually distributing flyers in the school and asking students to vote for her. But the night is finally here, and people have dressed up for the event. Tiffany is wearing a beautiful blue and black gown, while Lori is wearing a white dress.

While Lori and Tiffany try to prove who is better, something sinister is taking place within the walls of Shadyside High. Linda and her prom date, Bobby, enter a secluded area where Linda sees that her flyers have been torn down. They suddenly hear the door of the room getting locked, and Linda goes into the dark to see what's happening. Bobby gets nervous and hears Linda making her way back to him. However, her intestines are in her hands, and she is killed by the same masked man. However, Megan hears Debbie's screams and realizes that something is wrong. A few moments later, he kills Bobby brutally, too. Meanwhile, Debbie and Judd become the next victims. During that time, Tiffany meets Lori and talks about what actually happened between her mother and father, and how she might have killed him. By listening to how low Tiffany has stooped to win the crown, Melissa starts ignoring her. But that makes Lori realize that she needs to up her game and don a new look by applying makeup. After coming back to the event, she beats Tiffany in a dance-off. Moreover, Tyler, who is Tiffany's boyfriend, is also falling in love with Lori.

Megan, on the other hand, tells Lori that something is wrong, and all the women nominated for Prom Queens are disappearing. In the meantime, Melissa is killed by the killer, and it is revealed that there are actually two killers on the loose. While listening to Megan, Lori believes Megan doesn't want her to be normal and have a decent relationship or friendship with anyone. Megan is infuriated by hearing this and realizes that she has changed just because Tyler has shown how much he loves her. Megan leaves Lori and starts investigating what was happening. It doesn't take much time for Lori to see that Megan was right, as Tyler is killed right in front of her. On the other hand, Megan stumbles on the dead bodies of Debbie and others. Megan and Lori finally meet as they try to run away from the killer. They make their way to the event, and it is revealed that Lori has been chosen as this year's Prom Queen, but Lori wants the school to know about the killings. The killer makes his way to the event and is soon captured by the students. When the mask comes off, Tiffany's father turns out to be the killer. When asked why he did this, he said that Tiffany and her mother badly wanted to win.

After all that happened, the police wanted to get a statement from Tiffany's mother, Nancy Falconer. So, Lori helped Tiffany and accompanied her home. They spent the night together, but soon someone made an entrance into Tiffany's house with a huge knife in their hands. It turns out that the person was Nancy. She tried killing them, so Tiffany and Lori hid themselves in one of the cupboards. But soon it is revealed that it was all a part of the plan, and they, both Tiffany and Nancy, wanted to kill Lori. In the process, Nancy confessed that it was she who killed Lori's father because she loved him, and her mother took him away from her. She was inching closer to hitting her with a knife when Tiffany said that she wanted to kill her. But as soon as she tried to do that, Lori kicked her powerfully and threw her to the ground floor, killing her instantly. Nancy went into a rage and just wanted to kill her, but Lori got the better of her and struck her on the head. Nancy falls to the ground, and Lori finally makes it out of their home.

The film has a mid-credits scene, which reveals that Nancy is still alive and her blood forms a kind of pentagram on the floor, which is often used in magic or the occult, making it clear that this story is far from over.
