Unlike many other main characters in anime, Kazuma tends to be quite shameless, lacking many of the usual chivalrous qualities a hero is expected to have. In particular, he often talks about believing in the kind of gender equality that would allow him to drop kick a girl if he needed to.
Kazuma first says this line right after his duel with Kyoya Mitsurugi, during which he steals his sword with a surprise attack and knocks him out with one blow. Mitsurugi's followers chastise Kazuma, but then he threatens them with vague gestures while saying this line. While Kazuma only says this line once, he repeatedly references gender equality time and again throughout the series.

Kazuma's perversions don't seem to extend to the goddess, as he has no attraction to Aqua whatsoever, and while he does have some affection for Eris, that doesn't exactly translate into lust. He learns fairly early that Eris wears pads to boost her relatively small bust, but he isn't all that bothered by it. Kazuma is attracted to Eris because of her personality, and he even tells her that he doesn't care that she pads her chest after dying again in Season 2, shouting the line above.

Kazuma's complaints about Aqua are no surprise to any KonoSuba fan. Despite being a goddess, she is overly haughty and conceited, and frequently makes trouble to the point that she is more helpful when she is doing nothing. Even when she is being helpful, she gets so obnoxious about it that any gratitude Kazuma feels quickly evaporates. In one such situation, Kazuma was brought back to life by Aqua, but her self-aggrandizing attitude soured on Kazuma, leading him to ask for a change in goddesses.
Kazuma would say this again in Season 3 when his poor initial impression of Princess Iris led him to ask for a new princess. Though Kazuma would eventually come to appreciate and become friends with Iris, this quote was Kazuma's best way of being rude and dismissive shortly and succinctly.

As a low-level adventurer with average stats and a cowardly personality, Kazuma certainly has a lot of weaknesses. Of course, his biggest weakness is beautiful women, and at the beginning of the series, he even had a hard time dealing with Darkness when they first met. This is all due to his long life as a shut-in, secluded in his home where he was able to avoid any manner of social interaction, including ones with girls.

Komekko is more potent than first impressions dictate and could quite easily overtake Megumin to become the most powerful Crimson Demon in KonoSuba.
Never was this particular weakness more apparent than during the KonoSuba movie, when Kazuma faced off against the Devil King General Sylvia. Sylvia's voluptuous and statuesque figure instantly charmed Kazuma, and he immediately surrendered to her embrace without even attempting to resist. However, Sylvia was not just endowed with female charm, a fact Kazuma had to find out the hard way.

Initially a quick ad-lib by Kazuma's voice actor, Jun Fukushima, this particular quote has become an iconic running gag for the character, one that has persisted throughout the anime. Though it is an aspect of Kazuma not originally from the light novel, it complements his general personality so well that it's hard to imagine a version of him that doesn't say this quote from time to time.
In the anime itself, Kazuma usually says this line whenever somebody starts calling out his name frantically. For example, the first occurrence of this happens when Darkness gets cursed by the Devil King General Verdia. Darkness is consumed by the excitement of the situation, and Kazuma, having no idea how to respond to Darkness, simply responds to his name being called with this line. It's a line that demonstrates both his flippant nature and his exasperation, two key aspects of his personality.
