In Naruto, Kekkei Genkai are the rarest type of abilities. They cannot be obtained by training rigorously, as they are linked to a specific bloodline. In fact, many people have tried to obtain them through unethical means.

Hashirama Senju
Kekkei Genkai: Wood Release

Wood Release is, without a doubt, one of the rarest and most highly sought-after Kekkei Genkai in Naruto. The first person to wield the Kekkei Genkai was none other than Hashirama Senju, who was the first Hokage of the Hidden Leaf. Orochimaru and several others tried to get their hands on the Kekkei Genkai, but most failed. Out of Orochimaru's numerous human experiments, only Yamato was able to use Wood Release.
However, he couldn't utilize the Kekkei Genkai on the same level as Hashirama. The latter was so proficient at using Wood Release that he could make a giant wooden statue that dwarfed even the Nine Tails. He also used the Kekkei Genkai to take the tailed beasts, which he later distributed to other villages. Hashirama's Wood Release also enabled him to dominate and beat Madara Uchiha.
Nagato Uzumaki

The Rinnegan was the last of the Three Great Dojutsu to be introduced in the series, which makes sense as it is the strongest out of them. Before his death, he had the Dojutsu implanted into Nagato Uzumaki. As a member of the legendary Uzumaki clan, Nagato had enough chakra reserves to harness the power of the Rinnegan.

Haku was bestowed with the Ice Release, which allowed him to create different structures with ice. He could create ice shards that could pierce the body of an individual and cause serious injuries. Haku's famed mirror technique also made use of Ice Release. While Ice Release wasn't an overpowered ability, it certainly was rare.
Toneri Otsutsuki

Toneri Otsutsuki was the main antagonist of Naruto The Last, a movie set after the events of the Fourth Great Ninja War. Toneri was obsessed with power, which propelled him to steal Hanabi's Byakugan. Not long after the incident, Toneri awakened the Tenseigan.

Isshiki Otsutsuki possessed an extraordinary Dojutsu called the Kokugan. The Kekkei Genkai granted him access to Sukunahikona, a Shinjutsu with which he could shrink any inanimate object and himself. If Sukunahikona wasn't enough, the Kokugan vested Jigen with the Daikokuten, which is another insane ability. With Daikokuten, Isshiki could seal away anything that had been shrunk by Sukunahikona.
Kimimaro
Kekkei Genkai: Shikotsumyaku

Kimimaro was the strongest member of the Sound Five. He belonged to the Kaguya clan, which went extinct because of its reckless demeanor.
Since Kimimaro was a member of the Kaguya clan, he had the potential to awaken the Shikotsumyaku. Fortunately, he awakened the Kekkei Genkai, allowing him to manipulate his bones at will.
