Albert Wesker has long been considered the major overarching villain of the Resident Evil series, and while his menacing appearance is already terrifying enough, what also makes him so eerie is how willing he is to kill anyone he considers an enemy. Despite not appearing in the flesh until Resident Evil 5, Spencer is massively important to the wider lore of the series. As the head of the Wesker project, Spencer was in charge of watching over the Wesker children, including Albert, while they were still developing. By the time he appears in the fifth game, Spencer is an old man who's hooked up to a life support system; clearly in the twilight years of his life.
All he can do in this state is go on long tirades about how he deserves to be seen as a god, despite that no longer being a possibility for him. During his final speech, moments before Chris and Jill barge into his room, Wesker stabs Spencer through the chest after seeing no use in keeping him around for any longer, ensuring only he can be considered a true god with nobody to stand in his way.

In reality, everything had been planned by Wesker as a way to test the abilities of Umbrella's B.O.Ws, but one character who almost managed to reveal Wesker's true affiliations was Enrico.

Enrico may have eventually been injured by an unknown B.O.W after finding his way to the mansion, but it was actually Wesker who put an end to him once and for all by shooting him in the chest. Sergei Vladimir is a rather interesting character in the series who only shows up in the Umbrella Chronicles spin-off game. As someone who rose up Umbrella's ranks in barely any time at all, Sergei regularly came into contact with Wesker, whom he would come to distrust. Sergei suspected Wesker was the type of guy who would willingly turn on his comrades if he spotted an opportunity, which is something Sergei himself got to witness first hand after being brutally murdered by the superhuman.
After Sergei's own personal facility was raided, Wesker, who was now far faster and stronger than Sergei, was able to easily overpower Sergei, even after he mutated. Wesker won't let anyone stand above him in the pecking order, with Sergei being a particularly annoying thorn in his side for much of his career.

When Wesker and Birkin joined Umbrella, James Marcus slowly started to lose much of his recognition as a scientist. What really sealed his fate was when Birkin was officially credited with creating the T-virus, which brought much humiliation to Marcus. Eventually, Spencer determined that Marcus was simply inadequate to exist in the modern era of Umbrella, and as so, sent an order for him to be assassinated.

The assassination was a joint effort between Wesker, Birkin, and a few other grunt soldiers who succeeded in gutting Marcus from the company so that they could take Umbrella's top spots for themselves. Excella acts as a loyal companion to Wesker for much of Resident Evil 5, using her prestigious position in Tricell to help him in his mission of taking over the entire world with Uroboros. However, Excella clearly hadn't read up on who Wesker actually was before meeting him, considering she genuinely believed he cared for her. Inevitably, though, once Excella had served her purpose, Wesker decided to use her as a freakish example of how Uroboros can affect the host's body.
Chris and Sheva eventually stumble across Excella not long after they board Wesker's cruise ship. After screaming out Wesker's name one final time, Excella's body begins mutating into a gigantic monster which takes over the entire tanker. Excella would actually come to harbor intimate feelings for Wesker during their adventure, so the fact he was more than happy to sacrifice her, just to put on a show for Chris and Sheva, emphasizes how cruel he truly is.

At a young age, Lisa Trevor and her mother were kidnapped by Umbrella and used as guinea pigs for their wide array of handcrafted viruses and diseases. While Lisa's mother ended up losing her life, Lisa would become a grossly mutated monster who was forced to wander around the dark chasms of the Spencer Mansion, unable to do anything but scream out for her late mother.
In Umbrella Chronicles, fans get to see how Wesker managed to escape the mansion before it was destroyed, but not before he is encountered by none other than Lisa one final time. After pummeling her with a barrage of bullets, Wesker leaves her for dead after a chandelier falls on her body, with Wesker making one sickening final remark when he asks her to "stay dead this time". Wesker had absolutely no sympathy for the young girl that he turned into a monster, without a care in the world for the people whose lives he'd ruined.
